生存数据中样本量的计算(五)
首先,小胖给大家简单介绍一下我们比较常见的两组比较的,建立在proportional hazard ratio assumption基础上的一些比较简单的临床试验的样本量的计算方法。
1. Lachin and Foulkes Method
Lachin and Foulkes Method简称LF Method由Lachin 和Foulkes于1986年提出:
Lachin JM and Foulkes M. Evaluation of sample size and power for analyses of survival
with allowance for nonuniform patient entry, losses to follow-up, non-compliance, and
stratification. Biometrics 1986;42:507-19
LF Method适用于两组比较的maximum duration trial,它要求数据比较好地符合proportional hazard ratio assumption。
而关于LF Method的软件计算,我们一般可以使用PASS中的Logrank Tests(Lachin and Foulkes)模块来实现。
2. Kim & Tsiatis Method:
Kim & Tsiatis Method由Kim K 和 Tsiatis在1990年提出:
Kim K and Tsiatis AA. Study duration for clinical trials with survival response and early
stopping rule. Biometrics 1990(46): 81-92.
Kim & Tsiatis Method适用于两组比较的maximum duration/inforamtion trial,它要求数据比较好地符合proportional hazard ratio assumption。Kim & Tsiatis Method的一大优势便是它可以应用于中期分析。
关于Kim & Tsiatis Method的计算,主要使用East软件。
3. Rubinstein, Gail, and Santner Method:
Rubinstein, Gail, and Santner Method由Rubinstein, Gail和Santner于1981年提出:
Rubinstein LV, Gail MH, and Santner TJ. Planning the duration of a comparative clinical
trial with loss to follow-up and a period of continued observation. Journal of Chronic
Diseases 1981;34:469-79.
Rubinstein, Gail, and Santner Method与LF method类似,但公式计算更为简化些,可通过nQuery软件中的Test based on exponential survival, accrual period and dropouts模块
来实现。
